Effect of Bcl-2-associated athanogene 3 on the proliferation of esophageal cancer cells and its correlation with micro ribonucleic acid let-7

Abstract
ObjectiveTo investigate the effect of Bcl-2-associated athanogene 3 (BAG3) on proliferation of esophageal cancer cells and its correlation with micro ribonucleic acid let-7(miRNA let-7).
MethodsIn this study, the cancer tissues (observation group) and adjacent normal tissues (control group) were collected from 180 patients with esophageal cancer treated in the Second People’s Hospital of Jiaozuo from May 2016 to March 2019. The expressions of BAG3 and miRNA let-7 in the observation group and the control group were detected by real-time fluorescence quantitative polymerase chain reactio(PCR). The correlation between BAG3 and miRNA let-7 was determined by Pearson correlation analysis. EC109 and TE10 cells were cultured and transfected with overexpressed BAG3 and knocked down BAG3, as BAG3 overexpression group and the BAG3 knockdown group. Then the cell proliferation of the two groups was detected.
ResultsThe expression of BAG3 in the observation group was significantly higher than that in the control group (P<0.05). The miRNA let-7 in the observation group was significantly lower than that in the control group (P<0.05). BAG3 protein was negatively correlated with miRNA let-7. The proliferation rate of BAG3 overexpression group was higher than that of BAG3 knockdown group.
ConclusionsBAG3 is highly expressed in the cancer tissues and lowly expressed in adjacent normal tissues, indicating its promotion effect on esophageal cancer cells. Meanwhile, the BAG3 protein is negatively correlated with miRNA let-7, so knocking down the expression of BAG3 can inhibit the proliferation of cancer cells.
