Expression and function of forkhead box P3 in the tissue of rat gastric cancer model

Abstract
ObjectiveTo study the expression and function of forkhead box P3(Foxp3)in the tissue of rat gastric cancer model.
MethodsForty- six Wister rats were divided into two groups: group A(n= 23)given sarcosine ethyl ester hydrochloride and sodium nitrite carcinogen for 6 months to establish the rat gastric cancer model,and group B(n= 23)as the normal control group.The reverse transcriptase- polymerase chain reaction(RT- PCR), Western blotting,and immunohistochemistry were used to detect the expression and location of Foxp3 in the gastric tissues.Immunohistochemistry was used to detect the expression and localization of Foxp3 in gastric cancer tissues.The relationship between the expression of Foxp3 and gastric lesions was statistically analyzed.
ResultsThe expression of Foxp3 mRNA in the gastric tissue of group A existed to varying degrees, and increased with the grades, and there was no Foxp3 mRNA expression in the gastric tissues of group B.Immunohistochemistry revealed that the expression of Foxp3 protein was mainly distributed in the nuclei of gastric cancer cells.In 16 gastric cancer sections,4 cases of precancerous lesions,and 3 cases of proliferative lesions in group A, 10(62.50%), 2(50.00%) and 1 case(33.30%) was positive for the Foxp3 protein, respectively.In group B, no Foxp3 protein staining was detected all 23 cases.Statistical analysis showed a linear correlation of Foxp3 protein expression in gastric tissue and lesion classification(r= 4.26, P< 0.01).
ConclusionThe expression of Foxp3 universally exists in gastric cancer cells and tissues, suggesting Foxp3 may play important roles in the occurrence and development of gastric cancer.
