Slit homolog 2 promoter methylation sites expression and clinical significance in hepatocellular carcinoma

Abstract
ObjectiveDetected Slit homolog 2 (SLIT2) promoter CG sties methylation expression and characteristics in HCC tissues, to explore its clinical significance.
MethodsUse the total length of primer polymerase chain reaction (PCR) and DNA sequencing method for 40 cases of HCC carcinoma tissue and corresponding normal liver tissue, clear SLIT2 promoter CG sites changes of methylation.
ResultsIn SLIT2 promoter sequences from starting sites -570 bp to -550 bp, there have three sites (21, 22, 23) methylation are significantly higher (75.0%, 72.5% and 72.5%) than other CG sites (40.0%, 35.0%, 25.0%, 32.5%, 30.0%, 30.0%, 20.0%, 17.5%, 25.0%, 22.5%, 20.0%, 35.0%, 15.0%, 35.0%, 22.5%, 35.0%, 62.5%, 45.0%, 62.5%, 57.5%, 52.5%, 37.5%, P<0.05). SLIT2 promoter methylation is correlated with pathological grade, alpha fetal protein (AFP) and hepatitis B virus (HBV) infection (P<0.05), and no correlated with sex, age, liver cirrhosis, TNM stage, and capsular (P>0.05).
ConclusionSLIT2 promoter methylation is associated with the development and prognosis of HCC, and to detect the methylation site characteristics can be guide the targeted therapy of HCC and provides the theory basis for target selection in the future.
