Effects of scutellarin on autophagy of rat myocardial cells after hypoxia reoxygenation

Abstract
ObjectiveTo investigate the effects of scutellarin (SCU) on autophagy and the viability of cardiomyocytes H9C2 after hypoxia reoxygenation (HR) in rats.
MethodsThe study was divided into normal control group (group A), reoxygenation group (group B), 1 μmol/L SCU group (group C), 10 μmol/L SCU group (group D), 30 μmol/L SCU group (group E) and 100 μmol/L SCU group (group F). The ratio of microtubule-associated protein 1 light chain 3 (LC3)Ⅱ/LC3Ⅰ in each group was detected by Western blotting, and the viability of myocardial cells was tested by methyl thiazol tetrazolium (MTT) assay.
ResultsThe ratio of LC3Ⅱ/LC3Ⅰ in group B (6.623 30±0.482 32) was significantly higher than in group A (1.000 0±0.000 0; F=50.214, P=0.002). The ratio of LC3Ⅱ/LC3Ⅰ in groups C, D, E and F was lower than that in group B. The ratio of LC3Ⅱ/LC3Ⅰ in group E (2.150 00±0.303 48, P=0.012) and group F (2.236 70±0.170 10, P=0.018) was the lowest. The viability of the cells in group B [(49.86±1.40)%] was significantly lower than in group A [(93.47±1.48)%; F=44.645, P=0.001]. The viability of the cells groups C, D, E and F was higher than that in group B. The viability of cells in group E [(78.98±1.59)%, P=0.001] was the highest, and that in group F [(57.32±1.38)%] was the lowest (P=0.028).
ConclusionHypoxia 2 h and reoxygenation 12 h can obviously activate autophagy and cause HR injury. SCU can reduce myocardium HR injury by inhibiting autophagy, and the effect is the best when the concentration of SCU is 30 μmol/L.
