The correlation between myeloid-derived suppressor cells and regulatory T cells in lung cancer during perioperative period

Abstract
ObjectiveThis study aims to evaluate the changes of CD11b+ CD33+ human leukocyte antigen-DR (HLA-DR)--myeloid-derived suppressor cells (MDSCs) and CD4+ CD25+ forkhead box P3 (Foxp3)+ regulatory T cell (Treg) and to analyze the correlation between MDSCs and Treg in perioperative period in lung cancer patients.
MethodsNinety-eight lung cancer patients, aged 42-77 years, weighing 49-73 kg, were selected for radical resection through thoractomy. Peripheral blood was drawn at preoperation (T1), intraoperation (T2) and on 1st (T3), 3rd (T4) and 7th (T5) day after thoractomy. The concentration of MDSCs and Treg in peripheral blood mononuclear cells (PBMCs) was detected during perioperative period by flow cytometry (FCM) and automated hematology analyzer. The correlation between MDSCs and Treg was further analyzed at T1-T5. MDSCs and CD4+ cells isolated using FCM cell sorting and anti-CD4 magnetic beads were cocultured either in mixed pattern (without Transwell group) or separated using a Transwell chamber (with Transwell group). Five days after coculture, Treg cells were analyzed by FCM.
ResultsThe concentration of MDSCs at T3-T5 was significantly increased as compared with T1 [(0.684±0.326)×109/L, (0.545±0.307)×109/L, (0.496±0.302)×109/L and (0.219±0.133)×109/L, respectively, P=0.000、0.000 and 0.004]. The concentration of Treg at T5 was significantly increased as compared with T1 [(0.051±0.033)×109/L and (0.034±0.020)×109/L, P=0.006]. There was a positive correlation between MDSCs and Treg at T3-T5 (Pearson r=0.299 8, 0.434 2 and 0.477 3, P=0.004, 0.000 and 0.000, respectively). The number of Treg cells induced by MDSCs isolated at T3-T5 was increased as compared with T1 [(11.24±2.85)×104, (10.80±3.10)×104, (14.94±4.22)×104 and (6.71±2.56)×104,P=0.003, 0.000 and 0.000, respectively]. Compared with the "without Transwell group" , the absolute number of Treg induced by MDSCs isolated at T1-T5 was significantly decreased in the "with Transwell group" (P=0.008, 0.004, 0.000, 0.000 and 0.000).
ConclusionThere was a positive correlation between MDSCs and Treg during perioperative period in lung cancer patients. The capability of MDSCs isolated postoperatively was enhanced as compared to preoperative MDSCs and the induction of Treg was cell contact dependent.
