Expression and correlation of PRM1, PRM2 and TRF2 in idiopathic weak sperm disease

Abstract
ObjectiveTo study the mRNA and protein expression levels of PRM1, PRM2 and TRF2 in sperm of patients with idiopathic oligospermia and to explore its correlation with sperm motility.
MethodsForty-three patients with idiopathic oligozoospermia in the reproductive center of our hospital from January 2017 to December 2017 were selected as a research object. The sperm donors in our hospital were the control. The semen parameters of the two groups were compared, and the mRNA levels of PRM1, PRM2 and TRF2 were detected by RT-PCR. The protein level was detected by Western blot.
Results①The sperm density, sperm motility and normal sperm morphology were lower in the idiopathic oligozoospermia group than in the normal group(P<0.05). ②The relative expressions of PRM1 and PRM2 mRNA and protein in the idiopathic oligozoospermia group were significantly lower than those in the normal group(P<0.05), but the relative expression of TRF2 mRNA and protein was not statistically significant. ③There was a significant correlation between PRM1 mRNA and PRM2 mRNA and sperm forward movement(P<0.05).
ConclusionsPRM1 and PRM2 are related to the motility of male sperm and may be involved in the development of idiopathic oligospermia. The prevention and treatment of early idiopathic oligozoospermia can be carried out according to the expres-sion level of protamine PRM1 and PRM2, which provides molecular biological mechanism and reduces the occurrence of idiopathic oligospermia.
