Lymph node metastasis of tumor: the battle between tumor cells and immune system

Abstract
Tumor metastasis is one of the important reasons for poor prognosis of tumor patients, in which lymph node metastasis is the earliest and most frequent event. With the development of immunotherapy, tumor draining lymph nodes (TDLN), especially tumor-metastatic draining lymph nodes, as the key immune organ of anti-tumor immunity, has been paid more attention in the process of tumor development. After tumor metastasis, TDLN creates a special tumor immune microenvironment through various mechanisms, and this microenvironment may not only change the metabolism and other characteristics of tumor cells and then affect their ability of proliferation, migration and invasion, but also influence various kinds of immune cells in lymph nodes, leading to the emergence of immune tolerance and immunosuppression, which exactly responds to immune checkpoint blockade therapy. In this review, we discussed the regulatory effect of TDLN on tumor cells and immune system after tumor cell metastasis, and the new clinical definition and value of lymph node metastasis in the era of immunotherapy, which provides a new direction to refine immune therapy strategies.
