Genetic characteristics of 181 cases of mpox virus in Beijing in 2023

Abstract
ObjectiveTo analyze the genomic variations and genetic evolution characteristics of the mpox virus identified from May to September 2023 in Beijing, so as to provide the basis for development of mpox prevention and control measures.
MethodsSamples of throat swabs, skin lesion swabs, and anal swabs were collected from mpox cases. The amplicon sequencing method was use for viral genomes sequencing. The sequences were aligned and analyzed for variation. A phylogenetic tree and a haplotype network were constructed.
ResultsA total of 181 high-quality genome sequences of the mpox virus were obtained and 193 nucleotide mutation sites were identified. Among the mutation sites, 80.8% were driven by the APOBEC3 enzyme. Phylogenetical analysis results showed that the viruses belonged to the C.1 branch of the West African lineage and could be further divided into four different clusters. The haplotype analysis showed that 181 mpox virus genomes can be divided into 108 haplotypes.
ConclusionsThe current circulating mpox virus in Beijing is composed of multiple sources of importation along with continuous transmission and evolution of local mpox virus strains. It is necessary to continue genomic surveillance and research to evaluate the genetic variation of local viruses.
