Preparation and biosafety assessment of sustained-release drug-loading nano-micelle for the co-delivery of indocyanine green and paclitaxel

Abstract
ObjectiveTo analyze the nano-micelle drug-loading system for the co-delivery of indocyanine green and paclitaxel.
MethodsThe tri-block polymer of polyethylene glycol-polycaprolactone-polyethyleneimine(PEG-PCL-g-PEI) micelle was prepared and used for drug-loading; the paclitaxel was loaded by the self-assembled hydrophobic core of PCL; the indocyanine green was adsorbed by the positive charges on micelle surface. The cytotoxicity test, in vitro drug release test and near-infrared thermal imaging were used to verify the feasibility of drug-laoding and co-delivery. The routine blood test and biochemical test were used to evaluate the biosafety of polymer carrier.
ResultsThe drug-loading system was simultaneously loaded with indocyanine green and paclitaxel through physicochemical characteristics and methoxy-polyethylene glycol(MPEG)2 000-PCL6 000-PEI2 000had the best drug-loading capacity[(9.5±3.5)wt%] and entrapment efficiency[(87.2±4.4)%]. The co-delivery system showed tumor targeting effect through thermal imaging. The polymer micelle showed good biocompatibility through routine blood test and biochemical test.
ConclusionThe tri-block polymer of MPEG-PCL-g-PEI micelle can simultaneously load indocyanine green and paclitaxel, and it can effectively achieve the co-delivery of drugs with good biocompatibility.
