Expression changes and significance of F4/80, IL-6 and IGFBPrP1 in liver injury in beagles

Abstract
ObjectiveTo explore the expression changes of F4/80, interleukin-6(IL-6)and insulin-like growth factor binding protein-related protein 1(IGFBPrP1)in liver injury induced by thioacetamide(TAA)in beagles.
MethodsTwelve male beagles were randomly divided into two groups(n=6 in each group)using random number table method: normal group and TAA group(6 weeks). The morphological changes of liver tissues were observed.The expressions of F4/80 and IGFBPrP1 were detected by immunohistochemisty staining.The levers of IL-6 and IGFBPrP1 in plasma were detected by enzyme-linked immuno sorbent assay(ELISA). The protein expression of IGFBPrP1 was detected by Western blotting.
ResultsIn TAA group, hematoxylin-eosin(HE)staining results showed that there was hepatocyte degeneration, necrosis and infiltration of inflammatory cells.The results of immunohistochemisty staining showed that there were a large number of brown granules in Kupffer cells in TAA group, and the expression of F4/80 in TAA group was significantly increased compared with normal group(3.14±0.01 vs. 0.10±0.02, P<0.05). The plasma lever of IL-6 in TAA group was higher than that in normal group(26.00±2.90 vs. 16.8±4.10, P<0.05). The results of immunohistochemisty staining showed that there were a large number of brown granules in hepatic sinusoid, vascular wall, portal area and part of hepatic cells in TAA group, and the expression of IGFBPrP1 in TAA group was increased markedly compared with normal group(1.69±0.01 vs. 0.50±0.06, P<0.05). The Western blotting results showed that the lever of IGFBPrP1 in TAA group was higher than that in normal group(P<0.05). The ELISA results showed the plasma lever of IGFBPrP1 in TAA group was higher than that in normal group(1.54±0.08 vs. 1.09±0.05, P<0.05).
ConclusionThe expressions of F4/80, IL-6 and IGFBPrP1 change obviously in liver injury in beagles, which indicates all of them may involve in the process of liver injury.
