Experimental study on the mechanism of quercetin and luteolin-7-glucoside in promoting skin wound healing

Abstract
ObjectiveTo investigate the effect and mechanism of quercetin and luteolin-7-glucoside on re-epithelialization of skin wound.
MethodsThe full-thickness skin defect wounds of mice were treated with quercetin and luteolin-7-glucoside of different concentrations alone or jointly, and the wound healing was observed every other day. Hematoxylin eosin staining and immunohistochemical staining were used to observe the skin regeneration, cell proliferation and epidermal stem cell marker molecules β- Catenin, c-Myc, Sox2 expression. The expression of Sox2 in epidermal stem cells was detected by RT-PCR.
ResultsThe quercetin and luteolin-7-glucoside can promote skin wound healing, increase the thickness of new skin epidermis around the wound, increase the number of proliferating cells and epidermal stem cells in the epidermal basal layer β- Catenin, c-Myc and Sox2 expression. It can promote the expression of Sox2 in epidermal stem cells in vitro. The combination of quercetin and luteolin has an additive effect on wound healing.
ConclusionThe quercetin and luteolin-7-glucoside promote the proliferation of epidermal stem cells by upregulating the expression of β-catenin, c-Myc and Sox2, which in turn accelerates wound healing. The combination of quercetin and luteolin-7-glucoside has an additive effect on wound healing.
