目的探讨氢气对非小细胞肺癌的抑制作用及其机制。
方法将16只BALB/c雄性荷瘤裸鼠随机分为对照组和氢气组,氢气组给予吸入60%氢气+40%空气2 h/d处理。干预4周后结束实验,测量瘤体的体积及重量,采用苏木精-伊红染色法、免疫组织化学方法、实时定量聚合酶链式反应及Western blot检测肿瘤组织中Ki-67、环氧化酶2(COX-2)、血管内皮生长因子(VEGF)的含量。
结果与对照组相比,氢气组瘤体的体积及重量显著减小,氢气组Ki-67、COX-2、VEGF的mRNA、蛋白表达显著减少,且其病理形态及细胞异质性相对于对照组存在显著差异。
结论氢气干预可抑制非小细胞肺癌生长,其机制与抑制Ki-67、COX-2、VEGF表达有关。
ObjectiveTo investigate the inhibitory effect of hydrogen on non-small cell lung cancer and its mechanism.
MethodsSixteen BALB/c male tumor-bearing nude mice were randomly divided into control group and hydrogen group.Hydrogen group was treated for four weeks (60% hydrogen+ 40% air, two hours a day.The volume and weight of the tumor were measured.The levels of Ki-67, cyclooxygenase-2 (COX-2), and vascular endothelial growth factor (VEGF) in the tumor tissues were detected by hematoxylin-eosin staining, immunohistochemistry, real-time quantitative polymerase chain reaction and Western blot.
ResultsThe tumor volume and weight of the hydrogen group were significantly lower than those of the control group.The mRNA and protein contents of Ki-67, COX-2 and VEGF in the hydrogen group were significantly reduced, and its pathological morphology and cellular heterogeneity were significantly different from the control group.
ConclusionsHydrogen can inhibit non-small cell lung cancer, which is related to the repression of Ki-67, COX-2 and VEGF.
