目的通过网络药理学探讨甘草附子汤治疗类风湿关节炎(RA)的作用机制。
方法采用网络药理学方法,构建药物-靶点-疾病网络,进行富集分析,研究药物对靶点的作用、靶点的功能与疾病的相关关系,探讨甘草附子汤治疗RA涉及靶点和主要成分。
结果从中药系统药理学数据库与分析平台(TCMSP)中筛选出有效成分127个,得到49个甘草附子汤治疗RA潜在靶点,建立"药物-靶点-疾病"网络,得到135节点,473条边。筛选出Degree值较高的槲皮素、山柰酚、柚皮素等10个活性成分,以及白细胞介素(IL)-6、表皮生长因子受体(EGFR)、IL-1β、TP53、过氧化物酶体增生激活受体(PPARG)等30个Degree值较高的靶点。
结论甘草附子汤治疗RA主要与脂多糖反应、血管调节和血管反应、脂质与动脉粥样硬化、糖尿病晚期糖基化终末产物-晚期糖基化终末产物受体(AGE-RAGE)信号通路和辅助性T细胞(Th17)细胞分化等信号通路有关,为扩大甘草附子汤的临床应用提供理论依据。
ObjectiveTo explore the mechanism of Gancao Fuzi Tang in the treatment of rheumatoid arthritis (RA) through network pharmacology.
MethodsUsing network pharmacology methods, a drug-target-dis-ease network was constructed and enrichment analysis was performed to study the effect of the drug on the targets, the correlation between the function of the targets and the disease, in order to explore the targets and main compo-nents involved in the treatment of RA with Gancao Fuzi Tang.
ResultsA total of 127 effective ingredients were screened from the TCSMP database, and 49 potential targets for the treatment of RA with Gancao Fuzi Tang were identified. A "drug-target-disease" network was established with 135 nodes and 473 edges. A total of 10 active in-gredients with high degree values, including quercetin, kaempferol, and naringenin, as well as 30 targets with high degree values such as IL-6, EGFR, IL-1β, TP53, and PPARG were screened out.
ConclusionThe treatment of RA with Gancao Fuzi Tang is mainly related to the lipopolysaccharide response, vascular regulation and vascular response, lipid and atherosclerosis, diabetes AGE-RAGE signal pathway, Th17 cell differentiation and other signal pathways, which provides a theoretical basis for expanding the clinical application of Gancao Fuzi Tang.
